Is 5-HTP (5-Hydroxytryptophan) Safe with Tramadol?
Free · No account · based on published sources including NIH ODS, FDA drug labels, and peer-reviewed literature
Do not combine tramadol with 5-HTP supplements. If a patient is taking tramadol, advise them to stop 5-HTP immediately and consult their prescriber before restarting any serotonergic supplement.
Why this interaction occurs
Tramadol inhibits serotonin reuptake and 5-HTP is a direct serotonin precursor that increases serotonin synthesis. Combined use can cause excessive serotonergic activity leading to serotonin syndrome, a potentially life-threatening toxicity. SERT-deficient individuals (mimicked by SSRIs) are at even greater risk.
Evidence
Evidence level: moderate.
